You cannot add bike racks to the back of a cargo carrier. I would recommend the # CC-100 carrier and then add the # CC-125 bike adapter to it.

If the crossbars on your roof are long enough, and the total weight of the bikes you're carrying and the weight of the bike carriers don't exceed the weight capacity of the vehicle roof or the roof rack, you can certainly carry more than one bike.

We do have some options that will allow you to carry cargo and bikes on your 2012 Toyota Highlander, however, bike racks should not be mounted to a hitch mounted cargo carrier. The opening that you see on this cargo carrier is simply a hollow tube, not a receiver.

If you check out the attached video at the 1:44 minute mark the installer points out the locking latch feature of the # INA389 bike rack. This feature on the rear tire hold will prevent access to the clamp lever from being able to be lifted and clamp opened when the rack is in use or in the stored position.
